I've recently "upgraded" from TB v68 to version 78.5.1 on macOS 10.14.6, and my "From" column keeps disappearing. See here:

This video shows how I right-click the column headers to select the From column, and it appears correctly. But then I select any other mailbox, and when I return to the original mailbox, the From column disappears about 1/10th of a second later.

How do I fix this? Thanks!

does it do the same if you restart with addons disabled from the help menu?

Unfortunately yes, it does have the same behavior in Safe mode, with add-ons disabled from the Help menu.

Open a folder that has the columns organized correctly and includes From, then click the icon at the right end of the columns, Apply columns to, choose the faulty folder.

I just tried this, and when I then select the "applied-to" folder, the From column is there for 1/10th of a second, and then it disappears again.
